Footage description: Shots of an electron micrograph of a tissue section from a rat cortex. Steven Rose introduces the programme. Shots of a simplified diagram showing the same area of cortex. Only essential element have been included. Commentary by Rose points them out. Rose discusses the importance of isolating cell types from tissue so that the dynamics of cellular life can be studied. Shots of Holgar Nyden demonstrating his technique of nerve tissue microdissection which he developed in the 1950s. Shot of a nerve cell separated by this method. Rose explains the shortcomings of this method - it cannot be used to obtain large quantities of material for study. Rose discusses and demonstrates his own technique for separating nerve cells from nerve tissue for laboratory examination. This method overcomes the yield problem. Layers of Debris, Glia, Neurones and Blood cells are centrifuged. Rose discusses possible methods for determining the purity of his cell samples obtained by his own method. Shot of a fixed and stained preparation of a cell suspension. Rose points out the various cells found there and discusses their physical characteristics. Rose prepares and examines a sample under the light microscope. Shot of the sample under magnification. Rose explains the need for staining samples and demonstrates by staining a sample with methyl blue. Shot of the stained preparation. A neurone can be seen. Rose explains and demonstrates various microscopic techniques for testing the purity of his preparation. Phase contrast, interference and dark field microscopy demonstrated. Rose discusses and demonstrates methods of determining the integrity of cell suspensions. Rose explains how the electron microscope can be used to determine the integrity of a cell sample. He demonstrates the technique for preparing a sample for EM inspection. Film shots of a cell sample being prepared for and examined under the electron microscope. Commentary explains what is being done. Shots of electron micrographs of rat neurone cells. Rose compares the validity of microscopic techniques with biochemical techniques for establishing cell sample integrity.
